Sectioning with Microtome

A series of sectioning paraffin-embedding tissues. These tissues attached to woody blocks should be sectioned by the use of a rotary microtome with a disposable stainless steel blade. Thinly continuous sections should be cut with a scalpel on a black board or other objects every appropriate sizes. At this time, be careful not to blow off these sections. Some appropriate-sized sections should be mount on distilled water on a slide glass coated with albumin-glycerin, and then should be warmed on a hot plate around 45 C. This manipulation would make these sections developed. After this development, water should be absorbed with a piece of paper wiper, and then these developed sections should be arranged quickly on the slide glass outside of the hot plate. After arranging, the section-mounted slide glass should be warmed and dried on the hot plate again for more than 24 hours in order to attach the sections to the slide glass completely. For a more efficient staining, 10 or more pieces of slide glasses (maximum of 15 pieces) should be stored at a time in a staining basket.
